Overview

PNC-27 is an experimental 32-amino-acid synthetic construct engineered by attaching a cell-penetrating leader domain to an HDM-2-binding sequence derived from the p53 tumor-suppressor protein. In laboratory settings, it is evaluated strictly as an investigational agent to study structural biology and cellular interaction mechanisms. It is not approved for therapeutic administration in humans. Preclinical research primarily focuses on how this chimeric structure interacts with anomalous membrane targets. By attaching to abnormally expressed HDM-2 on target cell surfaces, the peptide forms pores that lead to cell lysis, differentiating its pathway from traditional apoptosis. Despite exploratory findings in cell and rodent assays, regulatory agencies have issued explicit warnings highlighting that PNC-27 lacks human safety validation, is unapproved, and has previously faced safety risks including manufacturing contamination.
