ACE-031 1mg

Investigational ActRIIB-Fc decoy protein evaluated for myostatin inhibition, discontinued due to vascular adverse events.

Get new protocol alerts and research updates

Mechanism

Acts as an ActRIIB decoy receptor that traps myostatin and activin-family ligands, preventing them from binding natural receptors to relieve the biological brake on muscle enlargement.

Dosing

Clinical protocols were terminated, though laboratory measurement references begin with a nominal 100 mcg benchmark.

Reconstitution

Reconstituting a 1 mg vial with 1.0 mL of bacteriostatic water creates an experimental concentration of 1.0 mg/mL.

Storage

Store the intact or reconstituted solution refrigerated between 2°C and 8°C protected from light, avoiding freezing or agitation to preserve protein integrity.

Reconstitution Steps

  1. 1

Clean the rubber stoppers of both the ACE-031 vial and the bacteriostatic water with sterile alcohol pads and allow to air dry.

  2. 2

Draw 1.0 mL of bacteriostatic water into a sterile mixing syringe.

  3. 3

Insert the needle at a slight angle and allow the diluent to run gently down the inside glass wall to avoid foaming.

  4. 4

Gently rotate or roll the vial between your palms until the lyophilized cake is fully dissolved without shaking.

  5. 5

Visually inspect the solution for clarity and refrigerate immediately between 2°C and 8°C.

Overview

ACE-031, also known experimentally as ramatercept, represents an engineered soluble fusion protein composed of the extracellular domain of the activin receptor type IIB linked to an IgG1-Fc tail. Formulated initially to address severe degenerative muscle-wasting disorders, it serves as a circulating decoy receptor that sequesters negative growth factors such as myostatin and activins before they can bind cell-surface receptors. While early Phase 1 trials demonstrated clear increases in lean mass and thigh volume in healthy volunteers, subsequent clinical evaluation in pediatric Duchenne muscular dystrophy was terminated early due to vascular off-target manifestations, including epistaxis and telangiectasias. Because broad TGF-beta family ligand binding affects non-skeletal tissues, the therapeutic index proved unacceptable for commercial drug development. Today, ACE-031 is referenced strictly as an experimental tool in comparative myostatin pathway biology. Standard research-grade vials are produced solely for in vitro analysis or laboratory measurement conversions, carrying significant off-target warnings and no verified safety clearance for administration.
