FOXO4-DRI 10mg
FOXO4-DRI is an investigational D-retro-inverso peptide researched as a senolytic targeting p53 interactions in non-viable cells.
Mechanism
Competitively inhibits the FOXO4-p53 complex at the p53 TAD2 interface, freeing p53 to selectively trigger apoptotic cell death within senescent cellular populations.
Dosing
Standard laboratory protocols report an initial starting level of 250 mcg administered once daily.
Reconstitution
Reconstituting a 10 mg vial with 3.0 mL of bacteriostatic water achieves an approximate working concentration of 3.33 mg/mL (33.3 mcg per U-100 syringe unit).
Storage
Maintain lyophilized powder in cold storage at -20 °C, and keep the reconstituted solution refrigerated between 2 °C and 8 °C without refreezing.

Reconstitution Steps
- 1
Draw 3.0 mL of bacteriostatic water into a sterile mixing syringe.
- 2
Slowly introduce the diluent against the internal glass surface of the vial to prevent foaming.
- 3
Gently roll or swirl the vial until the lyophilized cake is fully dissolved without vigorous agitation.
- 4
Record the date and preparation concentration on the vial label and place it immediately into refrigerated storage at 2–8 °C away from light.

Overview
FOXO4-DRI, alternatively identified in preclinical literature as Proxofim, is an engineered D-retro-inverso peptide derived from the FOXO4 transcription factor sequence. Its synthetic design incorporates modified stereochemistry that grants resistance to enzymatic degradation, permitting extended stability within biological systems. The molecule has garnered significant scientific attention within anti-aging and senolytic research as a targeted pathway disruptor. Preclinical animal models indicate that FOXO4-DRI works by interfering with the intracellular binding between FOXO4 and the tumor suppressor protein p53. In senescent cells, this disruption releases p53 into the cytoplasm to initiate targeted apoptosis without damaging adjacent healthy, non-senescent tissues. Animal studies have observed improvements in physical performance, renal function markers, and coat density after senescent cell clearance. All existing investigations regarding FOXO4-DRI remain strictly confined to cellular assays and rodent models. It is an unapproved, experimental peptide without established clinical safety, toxicity profiles, or validated human pharmacokinetics, remaining solely designated for controlled laboratory experimentation.
