Dihexa 10mg
Synthetic angiotensin IV analog researched for neuroplasticity and HGF/c-Met-mediated synaptogenesis.
Mechanism
Dihexa binds with high affinity to hepatocyte growth factor (HGF), potentiating c-Met receptor phosphorylation and stimulating downstream cascades that drive spinogenesis and synaptogenesis.
Dosing
Research references document 5–10 mg, typically administered sublingually or orally.
Reconstitution
Add 2.0 mL bacteriostatic water to achieve a final working concentration of 5 mg/mL.
Storage
Store lyophilized peptide at −20 °C; once reconstituted, refrigerate at 2–8 °C and protect from light.

Reconstitution Steps
- 1
Remove the Dihexa 10mg lyophilized vial and bacteriostatic water from cold storage and allow both to equilibrate to room temperature for 15 to 20 minutes.
- 2
Disinfect the rubber stoppers of both vials using fresh, sterile alcohol swabs and allow them to air-dry completely.
- 3
Draw exactly 2.0 mL of bacteriostatic water into a sterile mixing syringe.
- 4
Insert the needle into the Dihexa vial and slowly dispense the diluent down the inner glass wall to minimize turbulent agitation.
- 5
Gently swirl and roll the vial between your palms until the powder is fully dissolved into a clear, particle-free solution without shaking.
- 6
Affix a label indicating the reconstitution date and 5 mg/mL concentration, and transfer the vial to 2–8 °C refrigeration for laboratory storage.
Supplies Needed

Peptide Vial
Provides 10 mg of lyophilized Dihexa research peptide, requiring one vial for every 1 to 2 experimental cycles at the documented 5 to 10 mg parameters.

Insulin Syringes (U-100)
Facilitates accurate aliquot measurement from the 5 mg/mL reconstituted stock, requiring one sterile syringe per scheduled research draw.

Bacteriostatic Water
Supplies the preserved solvent for reconstitution, consuming 2.0 mL per 10 mg vial.

Alcohol Swabs
Used to sanitize vial septa prior to diluent introduction and stock withdrawals, requiring two to three swabs per protocol procedure.

Overview
Dihexa (N-hexanoic-Tyr-Ile-(6)-aminohexanoic amide, also identified as PNB-0408) is a synthetic oligopeptide derivative of angiotensin IV engineered to address the rapid metabolic degradation typical of native peptide ligands. Designed for enhanced blood-brain barrier permeability and chemical stability, Dihexa serves as a primary reference molecule in preclinical investigations targeting neurotrophic activity, dendritic arborization, and synaptic plasticity. Preclinical studies indicate that Dihexa interacts with hepatocyte growth factor (HGF) to augment c-Met receptor phosphorylation and drive downstream signaling pathways, such as PI3K/Akt, in the central nervous system. In vitro and in vivo models demonstrate that this signaling facilitation can induce robust dendritic spinogenesis and synaptogenesis at picomolar concentrations. While extensively evaluated in preclinical models of cognitive impairment, neurodegeneration, and chemically induced amnesia, Dihexa remains an investigational research compound that is strictly intended for laboratory experimentation and is not approved for clinical diagnostic or therapeutic use.
